Title: Regenerasi dan Uji Respon Toleransi terhadap NaCl pada Gandum (Triticum aestivum L.) Galur R-036 dan R-040 = Regeneration and NaCl Tolerance Response Testing of R-036 and R-040 Wheat Lines (Triticum aestivum L.)

Abstract: Keberhasilan meregenerasikan embrio somatik hasil induksi variasi somaklonal dapat menunjang program pemuliaan tanaman.Tujuan dari penelitian ini adalah mengetahui kemampuan regenerasi embrio somatik gandum hasil induksi variasi somaklonal dan menguji respons plantlet gandum galur R-036 dan R-040 varian somaklonal terhadap salinitas.Pada penelitian regenerasi eksplan yang digunakan adalah embrio somatik primer (ESP), embrio somatik kuartener (ESK) dan embrio somatik varian somaklonal (ESVS). Masing-masing eksplan diregenerasikan pada media 6mg/l BAP + 0,1mg/l IAA + 0,2 g/l casein hydrolyzate dan media 10mg/l Kin + 0,1mg/l NAA. Pada penelitian uji respons, plantlet dikultur pada tiga konsentrasi NaCl yaitu 0 mM, 150 mM dan 250 mM. Hasil penelitian menunjukkan bahwa komposisi ZPT yang digunakan pada masing-masing eksplan dari gandum galur R-036 dan R-040 tidak berpengaruh terhadap pembentukan plantlet, pucuk dan akar sedangkan pada eksplan ESP, ES bintik hijau diamati cenderung lebih tinggi dibandingkan ESK dan ESVS pada kedua media; plantlet hanya terbentuk dari eksplan ESP galur R-036 pada media 6mg/l BAP + 0,2 g/lcasein hydrolyzate + 0,1mg/l IAA dan galur R-040 pada media 10mg/l Kin + 0,1NAA mg/l. Respons toleransi salinitas plantlet yang berasal dari ESP galur R-036 dan galur R-040 pada konsentrasi 150 mM NaCl adalah “Peka” (P) dan pada konsentrasi 250 mM NaCl adalah “Sangat Peka” (SP).
The successful of somatic embryos regeneration from somaclonal-induced variants can support the plant breeding programs of wheat. The purposes of this research are to know the wheat regeneration abilities of somatic embryos from somaclonal-induced variants and to know the tolerance responses of wheat plantlets from somaclonal variants R-036 and R-040 lines, toward salinity. In this research, explants that was used in regeneration is primary somatic embryos (PSE), quaternary somatic embryos (QSE) and variants somaclonal somatic embryos (VSSE). Each explants has been regenerated on medium contain 6 mg/l BAP + 0,1 mg/l IAA + 0,2 g/l casein hydrolyzate and on medium with 10 mg/l Kin + 0,1 mg/l NAA. In the salinity testing responses, plantlets from regeneration was cultured on three different concentration of NaCl (0 mM, 150 mM dan 250 mM). The result show, PGR that was used in the medium of regeneration from wheat R-036 and R-040 lines has no effect in plantlet formation, neither shoot and root formation, whereas the explants from PSE can produce green spot significantly higher on the both regeneration medium than explants from QSE and VSSE; plantlets only produce of PSE explants from R-036 line on medium 6 mg/l BAP + 0,1 mg/l IAA 0,2 g/l casein hydrolyzate and PSE explants from R-040 line on medium 10 mg/l Kin + 0,1 mg/l NAA. The salinity tolerance responses of plantlets of PSE explants of R-036 and R-040 lines on medium contain 150 mM NaCl was “Sensitive” and on medium contain 250 mM NaCl was “Very Sensitive”
