Abstract: Modifikasi lipid dalam darah memegang peran penting pembentukan LDL teroksidasi yang akan terakumulasi sebagai sel busa yang berujung pada aterosklerosis dan penyakit jantung. Lobi-lobi (Flacourti inermis Roxb.) merupakan salah satu sumber daya alam yang mengandung antosianin dan dapat menghambat oksidasi lipid. Tujuan penelitian ini adalah untuk menentukan kandungan antosianin dan aktivitas antioksidan buah lobi-lobi, dan menentukan pengaruh asupannya (variasi dosis:13.975 mg/kg BB; 27.95 mg/kg BB; 41.925 mg/kg BB; and 51.90 mg/kg BB) terhadap konsentrasi kolesterol total secara in vivo. Aktivitas antioksidan ditentukan menurut metode kemampuan mereduksi dan penangkapan radikal DPPH. Sedangkan penentuan kandungan antosianin total digunakan metode diferensial pH dalam ekstrak metanol-HCl 1%. Antosianin total yang terdapat pada lobi-lobi adalah 141.27 ± 0.199 mg/100 g. Ekstrak metanol menunjukkan aktivitas antioksidan yang sedang pada pengujian penangkapan radikal DPPH dengan nilai IC50 sebesar 67.6 µg/L dan 0.0179 ± 0.001 K4Fe(CN)6 mek/g berdasarkan uji kemampuan mereduksi.Asupan lobi-lobi pada dosis 55.90 mg/kg BB selama 3 minggu secara kasar menghasilkan persen penurunan konsentrasi kolesterol total pada mencit (70.78 %).
Modification of blood lipids of low density lipoprotein (LDL) play a crucial role of oxidized LDL that accumulated as foam cell which can lead to atherosclerosis and heart disease. Batoko plum fruit (Flacourtia inermis Roxb.) is one of anthocyanin containing natural resources of East Indonesia that can inhibit lipid oxidation. The aims of this research were to determine anthocyanin content and antioxidant activity of batoko plum, and determine the effect of its intake (with various of doses: 13.75 mg.kg-1 of weights; 27.95 mg.kg-1 of weights; 41.925 mg.kg-1 of weights; and 55.90 mg.kg-1 of weights) of the total cholesterol based on in vivo experiment. Antioxidant activity of F. inermis was determined by reducing power and DPPH radical scavenging methods. Whereas, the pH differential method for determine the content of anthocyanin which its extraction method is maceration by using methanol-HCl 0,01%. Total anthocyanin were present in lobi-lobi at 141.27 ± 0.199 mg.100 g-1. Methanolic extracts showed the moderate antioxidant activity in the DPPH radical scavenging assay with IC50 value of 67.6 µg.L-1and 0.0179 ± 0.001 K4Fe(CN)6 Eq.g-1based on reducing power assay. Intake of batoko plumdietat dose of 55.90 mg.kg-1 of weights for 3 weeks widely resulted the highest percentage of decreases in total cholesterol concentration of mice (70.78 %).
