Abstract: | Tujuan penelitian ini adalah: Pertama, mensintesa kerabang telur ayam kampung dan kerabang telur ayam broiler menjadi biokeramik Hidroksiapatit (HAp). Kedua menentukan kadar kalsium - fosfat yang terkandung dalam biokeramik HAp. Ketiga mengkarakterisasi biokeramik HAp. Keempat menentukan aktivitas antibakteri HAp dengan kandungan kalsium dan fosfat tertinggi. Sintesa kerabang telur ayam dilakukan menggunakan metode presipitasi dengan suhu sintering 1000 ºC. Penentuan kadar kalsium – fosfat dalam HAp diukur menggunakan AAS dan Spektrofotometer UV – Vis. Karakterisasi senyawa HAP telah dilakukan dengan menggunakan FTIR. Penentuan aktivitas antibakteri dilakukan dengan menggunakan metode cakram kertas. Data hasil penentuan aktivitas antibakteri dianalisis menggunakan Rancangan Acak Kelompok (RAK) dengan 4 perlakuan dan 6 ulangan. Sebagai perlakuan adalah variasi konsentrasi HAp 5μg/μL; 25μg/μL; 50μg/μL; dan 75μg/μL, dan sebagai kelompok adalah waktu analisis.
Hasil penelitian menunjukkan bahwa 10 g serbuk kedua kerabang telur ayam menghasilkan serbuk HAp sebesar 88 % - 98 % w/w. Kadar kalsium dan fosfat kerabang telur ayam kampung dan broiler berturut – turut: 72,23 % dan 78,62 % ; 67,22 % dan 73,75 %. Hasil karakterisasi HAp dari kedua kerabang menunjukkan adanya gugus OH dan PO4 yang menunjukkan bahwa HAp hasil sintesa adalah benar senyawa HAp. Uji antibakteri HAp terhadap bakteri karies Lactobacillus acidophilus menunjukkan hasil yang positif baik pada dosis uji terendah (5μg/μL) maupun dosis uji tertinggi (75μg/μL), ditandai dengan terbentuknya daerah daya hambat sebesar 22,93±0,066 mm dan 33,08±0,031 mm (kategori kuat yaitu >20 mm). The aims of this study are: First, to synthesize local chicken eggshell and broiler chicken eggshell become bioceramics hydroxyapatite (HAp). Second, to determine the levels of calcium - phosphate contained in HAp bioceramics. Third, to characterizes the bioceramics of hydroxyapatite (HAp). Fourth, to determine the antibacterial activity of HAp with the highest calcium and phosphate content. The synthesis of chicken eggshell was performed using precipitation method with sintering temperature 1000 ºC. Determination of calcium-phosphate levels in HAp was measured using AAS and UV-Vis Spectrophotometers. Characterization of HAP compounds has been performed using FTIR. Determination of antibacterial activity was done by using paper disc method. Data on the determination of antibacterial activity were analyzed using Randomized Block Design (RBD) with 4 treatments and 6 replications. As the treatment is the variation of HAp concentration 5μg / μL; 25μg / μL; 50μg / μL; And 75μg / μL, and as a group is the time of analysis. The results showed that 10 g of each eggshell powder yielded HAp powder of 88% - 98% w/w. Calcium and phosphate levels of local chicken eggshell and chicken broiler eggshell respectively: 72,23% and 78,62% ; 67,22% and 73,75 %. The results of HAp characterization of both eggshells shows the presence of OH and PO4 groups indicating that the HAp of the synthesis is truely of the HAp compound. HAp antibacterial test against caries bacteria Lactobacillus acidophilus showed positive results both at lowest test dose (5 μg / μL) and highest test dose (75 μg / μL), characterized by the formation of diameter inhibitory area 22.93 ± 0.066 mm and 33.08 ± 0.031 mm inhibition (strong category > 20 mm). |
