Abstract: | Tanaman karet sangat rentan terhadap serangan penyakit Jamur Akar Putih (JAP) (Rigidoporus microporus). JAP dapat menyebabkan kematian pada tanaman karet sehingga serangan patogen akan menyebabkan kerugian pada produksi kebun. Untuk dapat mengetahui karakter patogen JAP maka dilakukan penelitian yang bertujuan untuk mengidentifikasi karakterisasi JAP. Karakter yang diidentifikasi yaitu laju pertumbuhan dan morfologi koloni. Penelitian ini dilakukan mulai bulan April-Agustus 2016 di Laboratorium CARC (Carotenoid Antioxidant Research Center) Magister Biologi UKSW Salatiga. Sampel yang digunakan yaitu 12 isolat jamur R. microporus. Semua sampel berasal dari perkebunan karet Merbuh dan Blimbing yang telah diisolasi. 12 isolat tersebut kemudian ditumbuhkan pada medium PDA (Potato Dextrose Agar). Pengamatan dilakukan setiap 12 jam dan 24 jam. Hasil yang diperoleh menunjukkan bahwa laju pertumbuhan masing-masing isolat bervariasi. Laju pertumbuhan tercepat berturut-turut yaitu BB6, BW5, MB4, MA3, KB1, KB2, MA2, KB3, MB5, MA1, KB4, dan BW7. Laju pertumbuhan koloni isolat BB6 (Blimbing) menunjukkan pertumbuhan yang tercepat. Dari hasil pengamatan, mula-mula hifa dari masing-masing isolat berwarna putih seperti kapas dan kasar, namun setelah beberapa hari menjadi lebih halus dan berwarna putih seperti wol. Rubber plants are very vulnerable to attack of White Root Fungus (JAP) (Rigidoporus microporus). JAP can cause death in rubber plants so that the pathogen attack will cause a loss on the production of the garden. To be able to know the character of pathog JAP hence conducted research which aim to identify characterization of JAP. The identified characters are the growth rate and the morphology of the colony. This research was conducted from April to August 2016 at the CARC Laboratory (Carotenoid Antioxidant Research Center) Master of Biology of SWCU Salatiga. The sample used was 12 isolates of R. microporus fungus. All samples were from isolated Merbuh and Blimbing rubber plantations. The 12 isolates were then grown on PDA medium (Potato Dextrose Agar). Observations are made every 12 hours and 24 hours. The results show that the growth rate of each isolate varies. The fastest growth rates are BB6, BW5, MB4, MA3, KB1, KB2, MA2, KB3, MB5, MA1, KB4 and BW7 respectively. The growth rate of isolate colonies of BB6 (Blimbing) showed the fastest growth. From the observations, initially the hyphae of each isolate are white as cotton and rough, but after a few days it becomes smoother and wool-like white. |
