Abstract: Tujuan dari penelitian ini adalah mengetahui pengaruh berbagai konsentrasi Penicillum chrysogenum (4%; 8%; 14%; 16%; 20%; 24%) terhadap produksi dan karakterisasi ekstrak kasar enzim pektinase dari limbah kulit nanas. Parameter yang digunakan dalam penelitian ini, untuk menentukan karakteristik ekstrak kasar enzim pektinase meliputi aktivitas enzim, suhu, pH, nilai KM, dan nilai Vmaks. Penlitian ini menggunakan rancangan percobaan RAK dengan uji lanjut BNJ. Hasil biomasa ekstrak kasar enzim pektinase yang terendah pada konsentrasi 4% dengan menghasilkan ratarata sebesar 1,63 gram dan hasil biomasa ekstrak kasar enzim pektinase tertinggi pada konsentrasi 24% dengan menghasilkan rata-rata 1,87 gram. Aktivitas enzim terendah pada perlakuan konsentrasi 4% dengan rata-rata sebesar 0,949 u/ml dan aktivitas enzim tertinggi pada perlakuan konsentrasi 24% dengan rata-rata sebesar 1,924 u/ml. Pada uji total padatan terlarut terendah pada perlakuan 4% dengan rata-rata sebesar 6,25 0Brix dan total padatan terlarut tertinggi pada konsentrasi 24% dengan rata-rata 8,273 0Brix. Enzim pektinase dapat bekerja secara optimum pada suhu 40 0C dengan pH 5,5 dengan berbagai konsentrasi. Hasil aktivitas enzim pada variasi suhu dan pH tertinggi, pada konsentrasi kapang 24% dengan suhu 40 0C dan pH 5,5. Analisis kinetika pektinase yang dihasilkan menunjukan nilai km yang terendah pada perlakuan konsentrasi kapang 24% dengan rata-rata 0,016 dan tertinggi pada konsentrasi kapang 4% dengan rata-rata sebesar 0,035. Nilai Vmaks paling rendah pada konsentrasi kapang 4% dengan rata-rata sebesar 0,474 dan nilai Vmaks tertinggi pada konsentrasi 24% dengan ratarata 0,690.
The purpose of this study was to determine the effect of various concentrations of Penicillium chrysogenum (4%; 8%; 14%; 16%; 20%; 24%) on the production and characterization of crude extract of pectinase enzyme from pineapple peel waste. The parameters used in this study, to determine the characteristics of the crude extract of the pectinase enzyme include enzyme activity, temperature, pH, KM value, and Vmax value. This study uses a RAK experimental design with a further BNJ test. The yield of the lowest crude extract biomass of the enzyme pectinase was at a concentration of 4% with an average yield of 1.63 grams and the highest yield of crude extract of the enzyme pectinase was at a concentration of 24% with an average yield of 1.87 grams. The lowest enzyme activity was at 4% concentration treatment with an average of 0.949 u/ml and the highest enzyme activity was at 24% concentration treatment with an average of 1.924 u/ml. In the test, the lowest total dissolved solids were at 4% treatment with an average of 6.25 0Brix and the highest total dissolved solids were at a concentration of 24% with an average of 8.273 0Brix. The pectinase enzyme can work optimally at a temperature of 40 0C with a pH of 5.5 with various concentrations. The results of the enzyme activity at the highest temperature and pH variations, at a mould concentration of 24% with a temperature of 40 0C and a pH of 5.5. The resulting pectinase kinetics analysis showed the lowest km value at 24% mould concentration treatment with an average of 0.016 and the highest at 4% mould concentration with an average of 0.035. The lowest Vmax value was at 4% mould concentration with an average of 0.474 and the highest Vmax value was at 24% concentration with an average of 0.690.
