Abstract: | Artemisinin adalah salah satu fitofarmaka penting yang dihasilkan oleh artemisia annua karena potensinya sebagai antimalaria, antikanker, antioksidan. Namun, produktifitas Artemisia annua L. di Indonesia secara umum masih berkisar di bawah 0,3% sehingga secara komersial belum layak untuk dibudidayakan. Peningkatan artemisinin dapat ditempuh melalui berbagai upaya untuk meningkatkan metabolit sekunder berupa artemisinin. Penelitian ini bertujuan 1) Mengetahui pengaruh konsentrasi 2,4-D dikombinasi dengan BA terhadap pertumbuhan dari eksplan Artemisia A. Linn. secara in vitro. 2) Memperoleh kombinasi perlakuan 2,4-D dan BA yang paling efektif dan efisien dalam meningkatkan kadar artemisnin 3) Memperoleh kombinasi perlakuan 2,4-D dan BA mampu menginduksi poliploid pada tanaman Artemisia annua Linn. Induksi poliploid secara kultur jaringan menggunakan zat pengatur tumbuh 2,4-D yang dikombinasi dengan Benzil-adenin (BA) pada konsentrasi 1,0; 1,5; 2,0 dan 3,0 mgL-1. Penelitian dilakukan pada bulan Juli 2022 hingga Juni 2023. Menggunakan Rancangan Acak Kelompok Faktorial dengan Analisis data pertumbuhan yang diperoleh menggunakan Analysis of Variance (ANOVA) dilanjutkan dengan uji DMRT. Hasil penelitian menunjukan bahwa perlakuan 2,4-D dikombinasikan dengan BA dengan lama penginduksian 21 mampu menginduksi poliploid A. annua Linn. Perlakuan BA dengan konsentrasi 1,5 mgL-1 kombinasi 2,4-D 2,0 mgL-1; Perlakuan BA dengan konsentrasi 2,0 mgL-1 pada 2,4-D konsentrasi 3,0 mgL-1; Perlakuan BA dengan konsentrasi 3,0 mgL-1 pada 2,4-D konsentrasi 2,0 mgL-1 merupakan kombinasi konsentrasi yang menghasilkan persentase tanaman poliploid tertinggi sedangkan untuk memperoleh kadar artemisinin pada perlakuan BA dengan konsentrasi 2,0 mgL-1 yang dikombinasikan dengan 2,4-D pada konsentrasi 1,5 mgL-1 kadar artemisinin dengan kadar artemisinin 0,473% (w/w) dan berat artemisinin 236,6 gram per tanaman. Untuk memperoleh kombinasi yang optimal dalam pertumbuhan eksplan terbaik pada konsentrasi BA dengan konsentrasi 3,0 mgL-1 yang dikombinasikan dengan 2,4-D pada konsentrasi 3,0 mgL-1 Artemisinin is one of the important phytopharmaceuticals produced by Artemisia annua due to its potential as antimalarial, anticancer, antioxidant. However, the productivity of Artemisia annua L. in Indonesia in general is still below 0.3%, so cultivating is not commercially feasible. Increasing artemisinin can be achieved through various efforts to increase secondary metabolites in the form of artemisinin. The aims of this study were: 1) Determine the effect of 2,4-D concentration combined with BA on the growth of Artemisia A. Linn. 2) Discover the most effective and efficient combination of 2,4-D and BA treatments in the artemisnin content 3) Obtaining a combination of 2,4-D and BA treatments capable of inducing polyploids in Artemisia annua Linn plants. Induction of polyploidy in tissue culture using growth regulator 2,4-D combined with Benzyl-adenine (BA) with concentrations of 1.0; 1.5; 2.0 and 3.0 mgL-1. The research was conducted from July 2022 to June 2023. The experiment were arraged in a Random Complete Block Design. Data were analysis of growth data obtained using Analysis of Variance (ANOVA) followed by DMRT test. The results showed that the treatment of 2,4-D combined with BA with an induction duration of 21 was able to induce polyploid A. annua Linn. BA treatment with a concentration of 1.5 mgL-1 combination of 2,4-D 2.0 mgL-1; BA treatment with a concentration of 2.0 mgL-1 on 2,4-D concentration of 3.0 mgL-1; BA treatment with a concentration of 3.0 mgL-1 at 2,4-D concentration of 2.0 mgL-1 is the optimal combination of concentrations because it produces the highest percentage of poliploid plants while to obtain the highest artemisinin content in BA treatment with a concentration of 2.0 mgL-1 combined with 2,4-D at a concentration of 1.5 mgL-1 artemisinin content with artemisinin content of 0.473% (w/w) and the weight of artemisinin 236,6 grams per plant. To obtain the optimal combination in the growth of the best explants at the concentration of BA with a concentration of 3.0 mgL-1 combined with 2,4-D at a concentration of 3.0 mgL-1. |
