Abstract: | Penggunaan pestisida sintetik tidak efektif mengurangi prevalensi penyakit karena menghasilkan residu dan tidak mendukung keanekaragaman hayati. Hal ini merupakan praktik yang tidak bijaksana dan dapat menimbulkan permasalahan di masa depan. Pemanfaatan mikroorganisme endofit banyak dilakukan untuk mengendalikan penyakit dan hama tanaman, karena dikenal memiliki mekanisme penghambatan patogen yang beragam. Agens hayati yang digunakan untuk menghambat patogen tanaman dan sudah banyak dikembangkan salah satunya adalah bakteri endofit. Tujuan penelitian yang dilakukan adalah untuk evaluasi potensi isolat bakteri endofit dari tanaman jahe sebagai agens pengendali hayati. Jenis penelitian yang dipakai deskriptif eksploratif. Pengambilan sampel tanaman jahe yang sehat dilakukan pada 5 titik di dua kecamatan yaitu Getasan dan Sumowono, Kabupaten Semarang. Isolasi dilakukan dengan mengambil tanaman jahe sehat dari bagian akar, rimpang, pelepah, dan daun. Selanjutnya karakterisasi dan seleksi isolat melalui pengujian kemampuan melarutkan fosfat, penambat nitrogen, aktivitas amilase dan protease, dan uji antagonis dengan Fusarium oxysporum secara in vitro. 20 isolat murni berhasil diisolasi terdiri dari 2 isolat yang berasal dari daun, 4 isolat dari pelepah, 8 isolat dari akar, dan 6 isolat dari rimpang. Hasil penelitian menunjukkan isolat D91 memiliki potensi sebagai agens hayati dengan mekanisme antagonis sebesar 86,91% didukung dengan aktivitas enzim protease, mampu melarutkan fosfat dan menambat nitrogen. The use of synthetic pesticides is not effective in reducing the prevalence of disease because it produces residues and does not support biodiversity. This is an unwise practice and can cause problems in the future. The use of endophytic microorganisms is widely used to control plant diseases and pests, because it is known to have various pathogen inhibition mechanisms. Biological agents that are used to inhibit plant pathogens and have been widely developed, one of which is endophytic bacteria. The purpose of the research was to evaluate the potential of endophytic bacterial isolate from ginger plants as biological control agents. The type of research used is descriptive and exploratory. Sampling of healthy ginger plants was carried out at 5 points in two sub-districts, namely Getasan and Sumowono, Semarang Regency. Isolation is carried out by taking healthy ginger plants from the roots, rhizomes, fronds, and leaves. Furthermore, the characterization and selection of isolates through testing phosphate solubility, nitrogen anchoring, amylase and protease activities, and antagonist tests with Fusarium oxysporum in vitro. 20 pure isolates were successfully isolated consisting of 2 isolates derived from leaves, 4 isolates from fronds, 8 isolates from roots, and 6 isolates from rhizomes. The results showed that D91 isolate has the potential as a biological agent with an antagonist mechanism of 86.91% supported by protease enzyme activity, able to dissolve phosphate and anchor nitrogen |
