Multiplication of leek (Allium porrum L.) by tissue culture is an alternative to the provision of seedlings, especially if the varieties no tillering and not able to flowering during plant production in Indonesia. A study was conducted to determine the best various of explant and growth regulator composition to induce embryogenic callus. Primary somatic embryos obtained by planted explant leaf base and root tip on MS medium containing 2.4 D and BA. At a later stage, primary somatic embryos will form a secondary somatic embryo after subculture. 1 month old root tips are the best explants for induce embryogenic callus, while ZPT 1.0 mg / l 2.4 D + 1.0 mg / l BA was the most appropriate composition to induce embryogenic callusen_US
