Abstract: An experiment regarding embriogenic calli induction and its proliferation in wheat seed explants on various auxin concentration and dark-light condition was carried out in Tissue Culture laboratory of Agricultural Faculty of Satya Wacana Christian University in Salatiga.The experiment was arrange in the Randomized Block Design, consist of three wheat genotypes (Dewata; R-039 and R-043); three 2,4-D concentration (2,5 ppm; 3,0 ppm; 3,5 ppm) and two condition of explants incubation (dark and light). Every combination of treatment were replicated three time. The purposes of the experiment were: 1. To obtain the concentration of 2,4-D which can induce the embriogenic callus and its proliferation from three genotypes of wheat seed explants. 2. To determine the incubation conditions which can support the callus induction and proliferation of three genotype of wheat seed explants. The Result of this experiment were : 1. The induction and proliferation of callus from three genotype wheat seed explants can be done through tissue culture using MS media enriched with minimum 2,5 ppm 2,4-D. 2. Both of dark and light incubation can support the induction of callus from three genotype wheat seed explants
