Abstract: Hasil dari induksi variasi somaklonal yang diperoleh perlu diregenerasikan dan di uji respon. Tujuan dari penelitian regenerasi adalah mengetahui pengaruh sorbitol terhadap regenerasi embrio somatik (ES) primer, ES kuartener (ESK) dan ES varian somaklonal (ESVS) dan mendapatkan komposisi media regenerasi yang mampu menghasilkan planlet pada genotipe R-039 dan Dewata. Penelitian uji respons bertujuan untuk mengetahui respons planlet gandum non varian somaklonal dan varian somaklonal. Dalam penelitian ini terdiri dari dua tahap, yaitu tahap regenerasi dan tahap uji respon yang keduanya menggunakan RAK dan analisis sidik ragam dengan uji BNJ 5%. Pada tahap regenerasi terdiri dari 12 perlakuan dan 9 ulangan. Perlakuan berupa 2 genotipe gandum, 3 macam eksplan dan 2 macam media. Tahap uji respons terdiri dari 6 perlakuan dan 14 ulangan, perlakuannya berupa planlet dari eksplan ESP yang dikultur di media dengan 3 konsentrasi NaCl (0 mM, 150 mM, dan 250 mM). Hasil penelitian menunjukkan sorbitol tidak berpengaruh nyata terhadap regenerasi planlet berdasarkan persentase ES yang telah berdiferensiasi menjadi planlet, pucuk, akar dan bintik hijau. Berdasarkan penelitian yang sudah dilakukan diperoleh planlet dan pucuk hanya di eksplan ESP dengan media I. Respons toleransi salinitas planlet gandum yang berasal dari ESP menunjukkan bahwa genotipe R-039 termasuk kategori “Sangat Peka” di media 150 mM NaCl dan “Sangat Peka” di media 250 mM NaCl, sedangkan Dewata termasuk kategori “Peka” di media 150 mM NaCl dan “Sangat Peka” di media 250 mM NaCl.
The result of the induction of somaclonal variation obtained needs to be regenerated and tested the response. The purpose of regeneration research was to determine the effect of sorbitol toward regeneration of primary somaic embryos, quartenary somatic embryos, and somatic embryos somaclonal variants get a regeneration medium composition capable of producing planlets on the genotype of R-030 and Dewata. The purpose of response testing to determine the response of wheat plantlets non somaclonal variants and somaclonal variants. This study consisted of two phases, namely the regeneration phase and the response test which both use RCBD and analysis of variance to test HSD 5%. The regeneration phase consisted of twelve treatments and nine replications. The treatment consist of two wheat genotypes, three kinds of explants, and two kinds of media (without and with sorbitol). Response test phase consisted of six treatments and fourteen replications, treatment in the form of ESP explants cultured in medium with three concentrations of NaCl (0 mM, 150 mM, and 250 mM). Result of research showed that sorbitol is no significant effect on the regeneration of plantlets by the percentage of somatic embryos which into plantlets, shoots, roots and green spots. Based on the research that has been done obtained plantlets and shoot only on explants of PSE with medium I. The response of the salinity tolerance of wheat plantlets derived from the ESP showed that genotype R - 039 are "Highly Sensitive" in the media 150 mM NaCl and "Highly Sensitive" in the media 250 mM NaCl , while Dewata are "Sensitive" in the media 150 mM NaCl and "Highly Sensitive" in the media 250 mM NaCl.
