Abstract: The objectives of the research are: Firstly, to determine the optimal yield of watermelon (Citrulus lanatus L) seed oil revealed by the length of extraction time. Secondly, to characterize the physico-chemical characteristic of watermelon seed oil. Thirdly, to identification of chemical composition of watermelon seed oil was done using Gas Chromatography-Mass Spectrometry (GC-MS). The extraction time had been done in between 10 until 20 hours using n-hexane as the solvent. Data of yield and physico-chemical of watermelon seed oil were analyzed by Randomized Completely Block Design (RCBD), 6 treatments and 4 replications. As the treatment is the extraction time which are (10; 12; 14; 16; 18; and 20 hours, respectively) and as the group is the time analysis. The result of the study shows that the length of time extraction of watermelon seed oil optimal at 18 hours is 16,79 ± 0,82 %. Long time extraction effect on yield, moisture content of oil, density, peroxide and saponification value. The contrary, there is no effect on the acid value of watermelon seed oil. GCMS analysis result, the watermelon seed oil ingredients content of 3 chemical components, they are 9,12-octadecadienoic acid 75,71%, hexadecanoic acid 18,10%, and octadecanoic acid 6,19%.
